ในสมัยอยุธยาปรากฏชื่อเมืองปราจีนบุรีเป็นครั้งแรก คำว่า "ปราจีนบุรี" เป็นคำสมาส เกิดจากคำว่า "ปราจีน" กับคำว่า "บุรี" คำว่า ปราจีนหรือปาจีน หมายความว่า ทิศตะวันออก ส่วนคำว่าบุรีหมายความว่าเมือง รวมแล้วคำว่า "ปราจีนบุรี" หมายถึงเมืองตะวันออก
